The ACELISA method involves several steps. First, a specific antibody that is capable of binding to the target antigen is immobilized onto a solid surface, often the wells of a microplate. Then, the sample containing the antigen is added to the wells and allowed to interact with the immobilized antibody. If the target antigen is present in the sample, it will bind to the antibody.
Next, a second antibody, known as a detection antibody, is added. This antibody recognizes a different part of the antigen and is linked to an enzyme. If the antigen is captured by the immobilized antibody, the detection antibody-enzyme complex will also bind to the antigen.
Finally, a substrate solution is added to the wells. The enzyme attached to the detection antibody catalyzes a reaction with the substrate, resulting in the development of a signal, often a color change. The intensity of the signal is directly proportional to the amount of antigen present in the sample.
ACELISA provides a quantitative measurement of antigens with high sensitivity and specificity.
